Abstract:
Background.Recentstudiesshowedthathyperglycaemia(HG)blocksanaesthetic-inducedpreconditioning.TheinfluenceofHGonanaesthetic-inducedpostconditioning(post)hasnotyetbeendetermined.Weinvestigatedwhethersevoflurane(Sevo)-inducedpostconditioningisblockedbyHGandwhethertheblockadecouldbereversedbyinhibitingthemitochondrialpermeabilitytransitionpore(mPTP)withcyclosporineA(CsA).
Methods.Chloralose-anaesthetizedrats(n?7–11pergroup)weresubjectedto25mincoronaryarteryocclusionfollowedbyminreperfusion.Postconditioningwasachievedbyadministrationof1or2MACsevofluraneforthefirst5minofearlyreperfusion.HGwasinducedbyinfusionofglucose50%(G50)for35min,starting5minbeforeischaemiaupto5minofreperfusion.CsA(5or10mgkg21)wasadministeredi.v.5minbeforetheonsetofreperfusion.Attheendoftheexperiments,heartswereexcisedforinfarctsize
measurements.
